Figure 2 Selection of cell lines. Upper panel: Stably transfected hEK-293 cells expressing the DProPHMs proteins indicated were rinsed with CSFM lacking bovine serum albumin and incubated in the same medium for 5 consecutive 24 h periods. Aliquots of (NH4 )2 SO4 pellets prepared from the spent media were fractionated by SDS-PAGE and visualized using rabbit polyclonal antiserum to rPAM(116-131) (Ab246). Apparent molecular masses are indicated. Lower panel: Duplicate aliquots of spent medium from DProPHM-382s, DProPHM-M109I, DProPHM-359s, and DProPHM-370s cells were fractionated by SDS-PAGE and visualized on immunoblots using antibody to rPAM(116-131) (Ab246) or to recombinant rPAM(37-382) (Ab475).
